Лептин, обладая всеми свойствами цитокина, продуцирует жировые клетки. Представляло интерес выявление соотношения концентраций лептина и активирующих превентивные реакции воспаления цитокинов в крови у практически здоровых северян при гиперлипопротеидемии.</p></sec><sec><title>Цель исследования</title><p>Цель исследования. Выявление соотношения концентраций лептина и IL-1ß, TNF-a, IL-6 и IL-10 в крови у практически здоровых северян при гиперлипопротеидемии.</p></sec><sec><title>Материалы и методы</title><p>Материалы и методы. Проведено обследование 286 практически здоровых людей в возрасте 35–55 лет, проживающих в Архангельской области. Индекс массы тела (ИМТ) у лиц первой группы (n=211) находился в интервале 19,5–26,8 кг/м² и соответствовал норме; и 75 человек второй группы ИМТ 31–40 кг/м²). В крови определяли концентрации лептина, интерлейкина-1β (IL-1β), фактора некроза опухоли альфа (TNF-a), интерлйкина-6 (IL-6) и интерлейкина-10 (IL-10) методом иммуноферментного анализа на автоматическом иммуноферментном анализаторе «Evolis» фирмы «Bio-Rad» (Германия), общего холестерола (ОХ), аполипопротеина А1 (АпоА-1), аполипопротеина В (АпоВ), липопротеидов высокой плотности (ЛПВП), липопротеидов низкой плотности (ЛПНП), липопротеидов очень низкой плотности (ЛПОНП), окисленных липопротеидов низкой плотности (оЛПНП), триглицеридов (ТГ), фосфолипидов (ФЛ) колориметрическим методом на биохимическом анализаторе «Stat fax 1904 Plus» фирмы «Awareness Technology, Inc.» США.</p></sec><sec><title>Результаты</title><p>Результаты. Гиперлипопротеидемия натощак у практически здоровых людей ассоциирована с повышением концентрации в крови лептина в пределах физиологического содержания. Установлена наиболее тесная взаимосвязь повышения уровня лептина и концентраций провоспалительных цитокинов IL-1ß, TNF-α и IL-6 (соответственно, r=0,68; 0,74 и 0,83; p&lt;0,001). Степень выраженности реакции провоспалительных цитокинов регулируется параллельным увеличением секреции противовоспалительного IL-10 (r=0,62; p&lt;0,001).</p></sec><sec><title>Заключение</title><p>Заключение. Следует признать, что стимуляция секреции лептином обеспечивается при влиянии более интенсивных сигналов, инициирующих цитокиновую реакцию. Для обеспечения секреции лептина, по сравнению с таковой реакцией провоспалительных цитокинов, требуется влияние более выраженных сигналов, ассоциированных с повышенной потребностью организма в метаболической энергии. Установлены более высокие концентрации лептина при низком содержании в плазме ЛПВП, лигандов АпоА-1 и АпоВ; обосновано мнение, что лептин в физиологических концентрациях регулирует использование депо энергетического субстрата жировой ткани увеличением его секреции при низком содержании в плазме ЛПВП, лигандов транспортных форм липидов.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>BACKGROUND</title><p>BACKGROUND: Cytokines secrete all cells when there is a threat of damage to it. Leptin, having all the properties of a cytokine, produces fat cells. It was of interest to identify the ratio of leptin concentrations and cytokines activating preventive inflammatory reactions in the blood of practically healthy Northerners with hyperlipoproteidemia.</p></sec><sec><title>AIM</title><p>AIM: To identify the ratio of concentrations of leptin and IL-1ß, TNF-α, IL-6 and IL-10 in the blood of practically healthy Northerners with hyperlipoproteidemia.</p></sec><sec><title>MATERIAL AND METHODS</title><p>MATERIAL AND METHODS: A survey of 286 practically healthy people aged 35-55 years living in the Arkhangelsk region was conducted. The body mass index in persons of the first group (n=211) was in the range of 19.5-26.8 kg/m2 and corresponded to the norm; and 75 people of the second group (body mass index 31-40 kg/m2). Concentrations of leptin, interleukin-1β (IL-1β), tumor necrosis factor alpha (TNF-α), interleukin-6 (IL-6) and interleukin-10 (IL-10) were determined in the blood by ­enzyme immunoassay on an automatic Evolis enzyme immunoassay analyzer from Bio-Rad (Germanytotal ­cholesterol (OH), apolipoprotein A-1 (ApoA-1), apolipoprotein B (ApoV), high-density lipoproteins (HDL), low-density lipoproteins (LDL), very low-density lipoproteins (VLDL), oxidized low-density lipoproteins (LDL), triglycerides (TG), phospholipids (FL) on the biochemical analyzer «Stat fax 1904 Plus» of the company «Awareness Technology, Inc.» USA.</p></sec><sec><title>RESULTS</title><p>RESULTS: Fasting hyperlipoproteidemia in practically healthy people is associated with an increase in the concentration of leptin in the blood within the physiological content. The closest relationship between an increase in leptin levels and concentrations of proinflammatory cytokines IL-1ß, TNF-α and IL-6 was established (r=0.68, 0.74 and 0.83, respectively; p&lt;0.001). The severity of the reaction of proinflammatory cytokines is regulated by a parallel increase in the secretion of anti-inflammatory IL-10 (r=0.62; p&lt;0.001).</p></sec><sec><title>CONCLUSION</title><p>CONCLUSION: It should be recognized that the stimulation of leptin secretion is provided by the influence of more intense signals initiating a cytokine reaction. In order to ensure the secretion of leptin, compared with that of proinflammatory cytokines, the influence of more pronounced signals associated with an increased need for metabolic energy is required.
