Ожирение считается неинфекционной пандемией, а рост его распространения представляет собой серьезную медико-социальную проблему. Высокие значения индекса массы тела тесно коррелируют с артериальной гипертензией и ее осложнениями, однако влияние ожирения на реализацию наследственной предрасположенности к гипертонической болезни (ГБ) остается слабо изученным.</p></sec><sec><title>Цель</title><p>Цель. Изучить ассоциации полиморфных локусов ММР с развитием ГБ у мужчин в зависимости от наличия ожирения.</p></sec><sec><title>Методы</title><p>Методы. Исследование проведено в дизайне «случай-контроль». В зависимости от наличия ожирения было сформировано две группы мужчин для исследования: с ожирением (больные ГБ – 323, контроль – 46) и без ожирения (больные ГБ – 241, контроль – 211). Всем мужчинам, включенным в исследование, проводилось генотипирование восьми полиморфных локусов MMP. Несинонимичные SNP выявляли при помощи онлайн-программы SIFT (<ext-link xlink:href="https://sift.bii.a-star.edu.sg/" ext-link-type="uri">https://sift.bii.a-star.edu.sg/</ext-link>). Функциональное значение полиморфных маркеров оценивали при помощи сервиса HaploReg (<ext-link xlink:href="https://pubs.broadinstitute.org/mammals/haploreg/haploreg.php" ext-link-type="uri">https://pubs.broadinstitute.org/mammals/haploreg/haploreg.php</ext-link>), влияние локусов на уровень экспрессии – с использованием GTEx-portal (<ext-link xlink:href="http://www.gtexportal.org" ext-link-type="uri">http://www.gtexportal.org</ext-link>).</p></sec><sec><title>Результаты</title><p>Результаты. Установлено, что у мужчин с ожирением в формирование ГБ вовлечены аллель А (OR=2,01; р=0,01) и генотип GG (OR=0,42; р=0,01) полиморфного локуса rs11568818 гена ММР7. У мужчин без ожирения с развитием заболевания ассоциированы аллель 6А (OR=1,32, р=0,04) rs3025058 гена MMР3 и генотипы GG (OR=1,52; р=0,04) и GА (OR=0,63; р=0,03) rs17577 гена MMР9. Показано, что полиморфные локусы, ассоциированные с развитием ГБ, характеризуются значимым регуляторным потенциалом – они располагаются в регионе гистонов, маркирующих промоторы и энхансеры, в области гиперчувствительности к ДНКазе-1, в сайтах связывания регуляторных белков и транскрипционных факторов, а также влияют на уровень экспрессии генов.</p></sec><sec><title>Заключение</title><p>Заключение. Таким образом, в настоящем исследовании установлены ассоциации с развитием ГБ полиморфного локуса rs11568818 гена ММР7 у мужчин с ожирением и локусов rs3025058 MMР3 и rs17577 MMР9 у мужчин без ожирения.</p></sec></abstract><trans-abstract xml:lang="en"><sec><title>BACKGROUND</title><p>BACKGROUND: Obesity is considered a non-infectious pandemic, and the increase in its spread is a serious medical and social problem. High values of body mass index closely correlate with arterial hypertension and its complications, but the effect of obesity on the realization of hereditary susceptibility to essential hypertension (EH) remains poorly understood.</p></sec><sec><title>AIMS</title><p>AIMS: To study the associations of polymorphic loci of MMPs with the development of EH in men depending on the presence of obesity.</p></sec><sec><title>MATERIALS AND METHODS</title><p>MATERIALS AND METHODS: The study was conducted in a case-control design. Surveyed 821 men – 564 patients with hypertension and 257 patients of the control group. Groups of patients and controls were divided into subgroups depending on the presence of obesity. All men were genotyped for eight polymorphic loci of MMPs. Nonsynonymous SNPs were detected using the software SIFT (<ext-link xlink:href="https://sift.bii.a-star.edu.sg/" ext-link-type="uri">https://sift.bii.a-star.edu.sg/</ext-link>). The regulatory potential was studied using the HaploReg service (<ext-link xlink:href="https://pubs.broadinstitute.org/mammals/haploreg/haploreg.php" ext-link-type="uri">https://pubs.broadinstitute.org/mammals/haploreg/haploreg.php</ext-link>). The association of SNPs with the expression level was detected using GTEx-portal (<ext-link xlink:href="http://www.gtexportal.org" ext-link-type="uri">http://www.gtexportal.org</ext-link>).</p></sec><sec><title>RESULTS</title><p>RESULTS: It was found that in obese men allele A (OR=2.01; p=0.01) and genotype GG (OR=0.42, p=0.01) of rs11568818 MMP7 are associated with the essential hypertension. In men without obesity allele 6A (OR=1.32; p=0.04) of rs3025058 MMР3 and genotypes GG (OR=1.52; p=0.04) and GA (OR=0.63; p=0.03)) of rs17577 MMP9 are associated with the development of the disease. These SNPs located in region of promoter and enhancer histone marks, in the region of hypersensitivity to DNAse-1, in binding sites of regulatory proteins and transcription factors.
